你 (you) + 说 (say) + 什么 (what). Literally 'you say what?' The question word 什么 asks for the content of what was said.
Use this when you didn't hear or understand what someone said. It's direct and can sound blunt, so use it with people you know. For a softer tone, you can add 对不起 (duìbuqǐ) before it. Avoid using it in formal settings; a more polite phrasing would be better.
